PrP^Sc can skip step 3 and directly infect the Peripheral Nervous System (PNS) or Central Nervous System (CNS) without involving the spleen.

Most likely the infectious agent goes from the spleen to the PNS and migrates to the spinal cord then to the CNS; migrating at about 1mm/day. The blood brain barrier may prevent direct infection of the brain.
